Hepatoprotective Activityof Holarrhena Antidysenterica Against Isoniazid and Rifampicin Induced Hepatotoxicity In Rats

Abstract
ABSTRACT: The present study investigates to find the hepatoprotective activity of leaves of Holarrhena antidysenterica. The extract of leaves of Holarrhena antidysenterica were screened for its hepatoprotective activity in isoniazid and rifampicin induced liver damage in Wister albino rats. The extract at dose of 300 mg/kg were administered orally once daily. Rats were treated with oral isoniazid (75mg/kg,) and co-administered with rifampicin (150 mg/kg,), daily for 15 days. Blood samples were collected from the heart at the 16th day. The degree of protection was measured by evaluating biochemical parameters such as serum glutamate Oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT), alkaline phosphatase (ALKP) and the level of total serum bilirubin. The livers collected were kept in formalin (10%) solution for the further histopathology study. The extract exhibited significant hepatoprotective effect and the biochemical parameters were restored towards normalization. Silymarin (25 mg/kg, po) was used as standard reference and exhibited significant hepatoprotective activity against isoniazid and rifampicin induced hepatotoxicity in rats. The protective effect of extract was also supported by histopathological study of liver tissues of animals. The results of this study strongly indicate that Holarrhena antidysenterica Linn have potent hepatoprotective action against isoniazid and rifampicin induced hepatic damage in rats.
